The southern terminus of S.R. 25 is at the junction with State Route 5 near Pine Hill in Wilcox County, and the northern terminus is at the Georgia state line in Cherokee County, approximately 20 miles east of Centre.

From Greensboro to its terminus, the orientation of S.R. 25 is in a general northeasterly/southwesterly direction. As the route begins its northeastward trajectory, it continues passing through rural areas and small towns, skirting the southern area of the Birmingham metropolitan area in Shelby County.
